ANA-12可選擇性直接與TrkB受體結(jié)合,抑制TrkB下游通路,而不改變TrkA和TrkC功能。在nnr5 PC12-TrkB細(xì)胞中,10 nM的 ANA-12可防止腦源性神經(jīng)營(yíng)養(yǎng)因子(BDNF)介導(dǎo)的神經(jīng)突增生。在DRG神經(jīng)元中,ANA-12可消除BDNF增加內(nèi)向電流的作用 |
體內(nèi)研究: |
在成年C57BL6/129SveV F1s小鼠中,ANA-12 (0.5 毫克/千克,腹腔注射)降低腦部TrkB活性,減輕焦慮和抑郁相關(guān)行為,而不影響神經(jīng)元的存活。在雄性C57BL/6小鼠體內(nèi),ANA-12 (0.5 毫克/千克,腹腔注射)對(duì)脂多糖誘導(dǎo)的抑郁行為顯示出抗抑郁作用。在雄性Sprague-Dawley大鼠中,ANA-12 (3微克/劑) 阻斷孤束內(nèi)側(cè)亞核(mNTS) BDNF減少進(jìn)食的作用。在雄性野生型小鼠中,ANA-12逆轉(zhuǎn)酒精攝入,下調(diào)D3受體表達(dá),但在D3R-/-型小鼠中是無(wú)效的。在雄性CocSired大鼠,腹腔注射ANA-12(0.5 毫克/千克)逆轉(zhuǎn)減弱的可卡因自身給藥 |
